Thymosin alpha 1 – the miracle peptide for immune health and longevity?

While you likely have heard of some of the more prominent peptides by now, including Ozempic, wegovy and mounjaro for their weightloss effects, and possibly also copper peptides for its skin benefits and BPC-157 for its purported gut healing, unless you are deep inside the biohacking or peptide world, thymosin alpha 1 might not have made it onto your radar (quite yet).

However, this might just be one of the more powerful (and for the majority likely safe!) peptides on the market. As part of my deep dive into the science, benefits (and potential risks!) of peptides, its potential benefits might just be life-altering (and extending).

Note: Most peptides are not currently approved to be safe for human use in most countries, including the UK, US and Europe. They can be purchased ‘for research purposes’, and due to their unregulated nature, there are many products on the market that might potentially be contaminated. If one was to experiment with peptides, make sure the provided peptides are third party tested for purity and efficacy, and ideally made in the US/ UK/ Europe.  This blog post summarises research and science for educational purposes, rather than protocols and medical information. But let’s get into. One of the peptides that really caught my attention, and the one we will discuss in today’s post, is thymosin alpha 1. Thymosin alpha 1 was first discovered in the 1970ies, so it has been around quite some time.

It naturally occurs in the thymus that has been recognized for modifying, enhancing, and restoring immune function. Thymosin alpha 1 has been utilized in the treatment of immunocompromised states and malignancies, as an enhancer of vaccine response, and as a means of curbing morbidity and mortality in sepsis and numerous infections (R).

Its synthetic, licensed version is the drug thymalfasin, is approved in more than 35 countries for the treatment of hepatitis B and C, and as an immune enhancer in several other diseases. Some of these include:  

  • Immune deficiencies
  • Psoriatic arthritis
  • Aging
  • Chemotherapy-induced toxicity
  • Cancer, in particular hepatocellular carcinoma, renal cell carcinoma, non-small cell lung cancer, melanoma, but also as ad adjuvant to other treatments in prostate (R, R), breast and other cancers (R)
  • Infections such as:
    • Human immunodeficiency virus (HIV), herpes viruses such as the cold sore virus, cytomegalovirus and Epstein Barr virus (R)
    • Pseudomonas
    • Mould toxicity and other fungal overgrowth
  • Sepsis
  • Acute pancreatitis (R)

Sounds too good to be true? And doesn’t the fact that in increases immune response also come with an inherent risk of it overstimulating the immune system, ie such as found in autoimmune conditions?

These were some of the questions I was asking myself. However, subsequent research into science papers showed opposing, quite astonishing, effects. Rather than running the risk of overstimulation, thymosin alpha 1 appears to be acting like a modulator, or ‘pleiotropic immunoregulator’ (R, R, R, R, R). It helps a weak immune system to become stronger, yet calms down an overreactive response!

It has been shown to have beneficial effects in calming inflammation and an angry immune response in conditions such as Multiple Sclerosis and other inflammatory and autoimmune conditions (R), potentially also allergies, histamine intolerance and Mast Cell Activation syndrome, by shifting the immune system from Th2 to a Th1 response. 

One of the problems associated with viral infections such as herpes infections is that their recurring reactivation is often associated with immune exhaustion, which impairs virus elimination (R, R, R, R).

Thymic peptides like thymosin alpha 1 enhance immune function and have been theorised to help reverse this exhaustion, in order to assist successful suppression of the virus.

Chronic viral infections such as Epstein Barr and cytomegalovirus have been shown to play a role in many disease processes, including cancers, chronic fatigue syndrome, Multiple Sclerosis, and more. Finding ways to target them could be potentially life-altering, improving quality of life and health span, and thereby promoting longevity at its core.

Concentrations of Thymosin alpha 1 are usually very high in fetuses and newborns, then after an initial drop should remain at a steady state level throughout adulthood. However, deregulated levels have been found in cancers, infectious diseases, chronic inflammatory autoimmune diseases such as psoriatic arthritis, rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE), with much lower levels that healthy controls (R).

The immune system and its role in longevity and health span

Aging is accompanied by remodeling of the immune system. With time, this leads to a decline in immune efficacy, resulting in increased vulnerability to infectious diseases, diminished responses to vaccination, and a susceptibility to age-related inflammatory diseases (R).

 

Research has identified 12 ‘hallmarks of aging’, which appear to be at the core of pretty much all age related decline and disease, including cardiovascular disease such as atherosclerosis, high blood pressure, strokes and heart attacks, neurodegeneration such as Alzheimer’s and Parkinson’s, type 2 diabetes, and the list goes on (R).

 

Rather than what used to be believed, that each disease has one distinct reason at its core, geroscience (the science of aging and longevity) found that it is in fact these 12 hallmarks that interconnect, like a cogwheel machine, affecting each other, and in totality contribute to the decline and ill health.

 

2 of these hallmarks are chronic low grade inflammation, also coined ‘inflammaging’, and the other ‘cellular senescence’, with the nickname of ‘zombie cells’. Chronic inflammation is like a slow gradual rusting of our cells, and senescent cells (=’zombie cells’) are cells that should have died and been replaced with new, fresh and healthy ones, but decided to linger. They in themselves secrete more inflammation, also called ‘SAPS’( = Senescence-Associated Secretory Phenotype).

 

Chronic infections and a gradual remodelling of the immune system contribute to both, chronic low grade inflammation, and immune senescence.

 

‘Fun fact’: Immunosenescence appears to develop earlier in men than women, as estrogen appears to have a immune increasing effect, and androgens (such as testosterone) an immune suppressing effect (R).

 

Obviously the first step in Functional Medicine and its root cause approach is always to look at the why. Why is there inflammation in the first place?

 

Are there chronic infections, hidden food intolerances, gut microbiome issues, a highly processed diet, lack of sleep and movement, environmental pollutants, mould from a water damaged building or stress that contribute to the inflammatory load?

 

But even despite best efforts to address those, some of my patients struggle to suppress for example latent viruses such as Epstein Barr infections, or their immune system is having a hard time dealing with mould and candida.

 

Thymosin alpha 1 might just be one of the missing tools in the toolbox towards optimal health and longevity that helps move the needle towards better health and quality of life, if all the other areas have been addressed (such as sleep, diet, movement, stress), yet the individual is struggling to get better.

 

In addition, due to its immune modulating, inflammation quenching effects, it might just help with slowing the ‘aging process’ (R). Early research, published last year, does point towards this being the case, with some of the most notable effects including:

  • Inhibition of ACE which supports blood pressure
  • Anti-atherosclerotic activity
  • Stimulation of neurogenesis, improving cognitive function (R)
  • And many more.
