
BPC 157 and the ‘wolverine repair stack, the secret to becoming invincible, or a potentially harmful hype?
If you have spent any time in biohacking, sports medicine, or longevity circles, chances are you have come across the peptide BPC-157.
Aging dramatically impairs tissue repair, contributing to increased recovery time from injuries, chronic non-healing wounds, and progressive musculoskeletal decline, with BPC-157 and TB-500 being hyped for counteracting this. These two peptides are often sold together as a ‘wolverine repair stack’, sometimes adding in a third peptide.
And while BPC-157 appears to have a mostly favourable and safe profile (read further to see where it may NOT be safe!), TB-500 might not. If one’s goal is not only to feel superhuman now, but also to stay healthy and not run into potential risks in future.

Today’s post looks at what research and safety data really says, and what might be overhyped (and potentially unsafe).

BPC-157
Let’s start with BPC-157
BPC- 157 is widely nicknamed the ‘Wolverine peptide’, inspired by the Marvel comic character Wolverine which possesses a range of superpowers including highly advanced self-healing abilities and a significantly prolonged lifespan.
And for good reason, as research on BPC-157 has shown some wonderful accelerated healing capabilities, most notably in the musculoskeletal system (ie muscles, tendons, ligaments) and gastrointestinal tract, but also, which is often less known, the cardiovascular system.
But first things first, what is BPC-157?
BPC-157 stands for Body Protection Compound-157. It’s a synthetic pentadecapeptide — a chain of 15 amino acids — originally isolated from human gastric juice by Dr. Predrag Sikiric in 1993 (R).
Animal studies have demonstrated regenerative and supportive properties across a remarkable range of tissues. BPC-157 is most notably known for repair and regeneration of:
- Tendons
- Ligaments
- Muscles
- Nerves
- Bones
- Teeth
- Increased athletic performance by improving recovery processes (R)
However, possibly even more so remarkable are its even wider reaching potential body wide benefits, including:
Brain health:
Cardiovascular health:
- Influence on blood clot formation and degradation
- Alleviated vascular occlusion disturbances (stroke) (R)
- Research suggests BPC 157 to be a potent blood vessel modulating agent, leading to optimization of the vascular response (R)
- Potent inhibitor of platelet aggregation and adhesion to the vascular wall (R)
- In heart disturbances, BPC 157 has been shown to assist in myocardial infarction, heart failure, pulmonary hypertension arrhythmias, and thrombosis prevention and reversal. The shared therapy effect occurred as part of its even larger cytoprotection (cardioprotection) therapy effect (direct epithelial cell protection; direct endothelium cell protection) (R).
Mental health:
- Brain function and mental health via normalising neurotransmitter signaling including dopamine, serotonin, GABA
- BPC-157 resulted in brain region-specific increases in serotonin synthesis. In addition, remarkably, it was found that BPC 157 effectively reduced some of the symptoms occurring in the serotonin syndrome, thereby modulating its levels rather than simply increasing it (R)
- Beneficial effects in neuropsychiatric conditions, such as depression (R).
And other beneficial effects including:
- Anti-inflammatory properties by reducing the inflammatory molecules IL6, TNF-alpha and IFN-gamma
- Highly protective against adverse effects by various medical agents, such as non-steroidal anti-inflammatory drugs (NSAIDs) and alcohol, particularly those related to liver and gastrointestinal tract damage (R)
- BPC 157 counteracted bleeding, thrombocytopenia, and in particular, leaky gut syndrome (R)
- Liver protection: showed a significant drop in bilirubin and aspartate aminotransferase. BPC-157 further also reduced the severity of fat accumulation in liver cells (R)
- Exhibits a strong antioxidant activity through its ability to stabilize free radical scavengers or counteract free radical formation and lesions. This, in turn, could testify in favor of the peptide’s suspected positive effect on the development of neurological diseases or even cancer, which have been known to be mainly caused by oxidative stress (R).
This all sounds like this might just be the magic cure for all?
Safety of BPC-157
Unfortunately, there have been limited clinical trials conducted in humans. To date, there have only been three published studies.
- Clinical pilot data derived from distinct populations indicated that 91.6% of elderly patients with chronic knee pain had significant improvement in knee pain (R), while
- 83.3% of middle-aged to elderly women with refractory interstitial cystitis experienced complete symptom resolution after a single intravesical dose (Lee et al., 2024). Although the exact mechanism of interstitial cystitis is unknown, there is thought to be a significant contribution from chronic inflammation, therefore BPC-157 was chosen in this study due to its anti-inflammatory properties and its role in restoring damaged epithelium as demonstrated in various animal models.
- A small 2025 trial showed no adverse effects on cardiac or renal function following intravenous administration of 20 mg BPC-157 (Lee and Burgess, 2025).
Possible side effects of BPC-157
Despite the favorable tolerance observed in these three studies, proposed side effects have been discussed based on the theoretical effects of BPC-157. These possible unwanted effects include pathologic angiogenesis (blood vessel growth), toxic metabolite formation, and overproduction of nitric oxide:
- Possible cancer risk with BPC-157: Pathologic angiogenesis (blood vessel growth) is implicated in the proliferation of tumor cells, as well as in immune and inflammatory disease processes (R). However, contrary to the theoretical tumor-promoting potential, BPC-157 has been shown in studies to inhibit uncontrolled cell proliferation, and in fact counteract tumorigenesis (R).
However due to the potential theoretical risk, and limited human safety data, the use of BPC 157 may not be the right choice in cancer patients until further human data confirms its safety (R).
- Overproduction of nitric oxide with BPC-157: BPC-157 robustly stimulates the nitric oxide system, which, at low levels, contributes to its cardiovascular health effects. However, at high levels, this has been shown to inhibit healthy red blood cell development and disrupt detox pathways, associated with the development of anemia and altered drug metabolism. Additionally, upregulation of nitric oxide has also been implicated in the development of neurodegenerative diseases such as Alzheimer’s, Parkinson’s disease and Multiple Sclerosis (R, R).
Research suggest that nitric oxide in moderate amounts can be neuroprotective while in large concentrations will be neurotoxic or cytotoxic.
BPC-157 might not be the right choice for women with endometriosis
And lastly, a third potential concern, which women with endometriosis might want to consider. This is still speculative in the context of BPC-157 specifically, but the mechanistic rationale is sound:
BPC 157 has been shown to be shifting from a pro-inflammatory profile (M1) towards a reparative profile (M2), which reduces fibrosis. However, in Endometriosis, M2 already appears to be overactive, so until further research confirms the opposite, I’d suggest staying clear of BPC-157 if you have endometriosis to avoid worsening of the situation (R).
Summarizing BPC-157
From looking at what research suggests thus far, for the majority of people, BPC-157 may be a wonderful tool from time to time.
Until further research has been conducted, I’d stay clear if you have a cancer history or endometriosis.
And be aware that more is not better, and too much nitric oxide production as stimulated by BPC-157 could backfire and damage red blood cells, brain cells and lower the body’s natural detox capacity.
However, the research paints a different picture for the other component of the Wolverine repair stack, TB-500.
TB-500
TB-500 is a synthetic version of Thymosin Beta-4. Studies suggest both may carry a potential cancer risk. Because TB-500 promotes new blood vessel formation (angiogenesis), cell migration, and tissue proliferation, it could accelerate the growth and spread of existing, undiagnosed tumors.
And while BPC-157 carries that same theoretical risk, the opposite has been shown for it in research thus far, with BPC-157 potentially inhibiting this growth and having cancer protective effects.
The same does not (currently) hold true for TB-500 and Thymosin Beta-4.
While TB-500 appears to help lower inflammation (R), its natural version, thymosin beta 4 has been shown to be overexpressed in human pancreatic cancer cells and to stimulate inflammation. Research suggests it might be involved in stimulating the progression of human pancreatic cancer (R), colon cancer (R), and has been shown to be associated with tumor metastasis (R).
However, also here it is not quite clear cut. Some conflicting research done in 2009 showed the opposite, with thymosin beta 4 demonstrating tumor suppressive effects, and its decreased levels resulting in poor prognosis and decreased survival in multiple myeloma (R).
I personally would likely stay away from TB-500 until more human research has been done to confirm their safety.
Even if they may make you feel invincible, more toned and ripped short term, are the potential risks worth it?
